The Molecular Biological Study for Anti-Cancer effects of Realgar on AGS |

ABSTRACT |
Background & Objective : Besides known effects of Korean traditional herbs, it was imperative to research for stronger anti-cancer drugs by conducting molecular biological studies. The Objective of this study was to find the effects of Realgar on AGS.
Materials & Methods : AGS from American type was used as culture. Realgar caused morphological changes and the following tests were conducted. 1. Tryphan bule exclusin test : to measure the killing effect on AGS. 2. MTT Assay : to Measure suppressive effect. 3. Quantitative RT-PCR : Revelation of Bcl-2, Bcl-XL, Bax and Fas. 4. Mitochondrial membrance potential and permeability transition.
Results: Realgar proved more positive effects than the controlled group. 1. Tryphan blue exclusin test : positive 2. MTT Assay : positive 3. Quantitative RT-PCR : decrease of Bcl-2 in proportion to concentration and increase of Fas. No change in Bax and Bcl- XL. 4. Realgar decreased the mitochondrial membrance potential.
Conclusion: Basic Realgar experiment proved its significant effect on AGS. However, more researches are needed to practically identify its effects. |
