Experimental Studies on Anti-cancer Effects of Soamgudamikgitang in Mice |

ABSTRACT |
Objective : A series of experimental studies were conducted to prove the antitumor effectiveness of Soamgudamikgitang.
Methods : Four-week-old C57BL/6 male mice were used in these studies. Experiments were conducted concerning the cytotoxic effect in vitro on B16-BL6 melanoma cells, the inhibitory effect in vitro on subfraction of DNA topoisomaerae I, the expression of metalloproteinase by RT-PCR method, the inhibiting effect in vitro on cell proliferation of BCE cells, the antiangiogenic effect in vitro in tube formation assay, the anti-angiogenic effect in vivo in chicken CAM assay, and the inhibitory effect in vivo on the metastasis of melanoma cell colonies.
Results: Concerning the cytotoxic effect in vitro, the IC50 of Soamgudamikgitang on B16-BL6 melanoma cells was 1,500 / . Concerning the inhibitory effect on the subfraction of DNA topoisomerase I, the effect of Soamgudamikgitang proved to be lower than that of camptotecin. Concerning the expression of metalloproteinase by RT-PCR method, the group treated with Soamgudamikgitang showed a general decrease in the expression of MMP2. However, the same group was also inclined to show a relative increase in the expression of TIMP1. Concerning in vitro cell proliferation of BCE cells, Soamgudamikgitang showed statistically significant(p<0.05) concentration-dependent inhibition effect on cell proliferation. Concerning tube formation assay, Soamgudamikgitang showed a concentration-dependent inhibitory effect. In the chicken CAM assay, the group treated with 1mg of Soamgudamikgitang showed high angiotoxicity. However, the group treated with 100㎍ of Soamgudamikgitang showed an 80% inhibitory effect. Concerning in vivo experimental lung metastasis assay, the groups treated with Soamgudamikgitang respectively showed a inhibitory effect.
Conclusions: The results of these experimental studies have proven the anti-metastatic effects of Soamgudamikgitang. Based on these results, our conclusion is that prescriptions containing Soamgudamikgitang will be highly effective in treating cancer patients. |
